The researcher is an experienced engineer or physicist with technical expertise in the field of microscopy hardware (optics, electronics, software). The candidate has a strong affection with biological research and has the ability to work in a team and also independently.
Confocal microscopy is a technology that makes 3D images of fluorescently labeled biological objects. Since the first development of the confocal microscope in the early 80s the detection efficiency has been improved substantially by using more sensitive detectors, better optical design and improvements of the acquisition electronics. It is the aim of this project to further improve the sensitivity of the microscope. To do that we will exploit the novel technology Controlled Light Exposure Microscopy (CLEM), invented and developed by Prof. E. Manders and Dr R. Hoebe. CLEM has been designed for reduction of photodamage in fluorescence live-cellmicroscopy. It is our aim to improve sensitivity by a factor of 50 to 100.
